In this talk I will discuss how we assess complement function in man and outline how we investigate complement deficiency states, emphasising genetic approaches. Deficiency states will be separated into two groups.

Firstly I will discuss deficiency of the activation proteins of complement. These deficiencies are associated with inability to mediate the important biological roles of complement. Not surpisingly recurrent infection is a common problem in this group of disorders. Secondly, I will discuss deficiency of complement regulators, These disorders are characterised by too much complement activation. The phenotypes are fascinating and have taught us much about the role of complement, particularly in the kidney.
